{"id":835,"date":"2023-04-12T05:00:43","date_gmt":"2023-04-12T05:00:43","guid":{"rendered":"https:\/\/anchrom.in\/blogs\/?p=835"},"modified":"2023-04-12T05:00:54","modified_gmt":"2023-04-12T05:00:54","slug":"hptlc-fingerprinting-analysis-of-cyperus-rotundus","status":"publish","type":"post","link":"https:\/\/anchrom.in\/blogs\/hptlc-fingerprinting-analysis-of-cyperus-rotundus\/","title":{"rendered":"TLC\/HPTLC Fingerprinting Analysis of Cyperus rotundus (Linn.)"},"content":{"rendered":"\n<h2 class=\"has-medium-font-size wp-block-heading\">What is HPTLC Fingerprinting ?<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\"><strong><u>High-Performance<\/u><\/strong><a href=\"https:\/\/anchrom.in\/hptlc\"><strong> Thin Layer Chromatography<\/strong><\/a> (HPTLC) is a modern version of TLC wherein the principle is the same but practice is different. HPTLC is <strong>GLP\/USP\/EP compliant technique<\/strong>.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">USP&#8217;s (United States Pharmacopoeia) definition of HPTLC fingerprint is \u2018It is the <strong>(electronic)<\/strong> image of the visual HPTLC chromatogram. Images are taken at 254nm, 366nm and in white light using suitable <a href=\"https:\/\/anchrom.in\/blogs\/tlc-hptlc-photodocumentation\/\"><strong>TLC photo docsystemstion system<\/strong><\/a> like <strong>CAMAG visualizer<\/strong>. Fingerprint is evaluated based on Rf, color and intensity of the separated bands.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">While developing fingerprint our aim should be to separate maximum number of compound with good resolution.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">For fingerprint development, <strong>BRM (Botanical Reference Material)<\/strong> and other related or confounding species are used for comparison. The same species of interest from different geographical or climatic conditions are also used to check variation in the samples.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\"><em>C. rotundus<\/em> is a traditional herbal medicine i.e. used for the treatment of pain, fever, diarrhea, dysentery, an emmenagogue and other intestinal problems. Mainly it is used to treat stomach ailments, wounds, boils and blisters etc.<\/p>\n\n\n\n<figure class=\"wp-block-image size-full\"><img loading=\"lazy\" decoding=\"async\" width=\"550\" height=\"246\" src=\"https:\/\/anchrom.in\/blogs\/wp-content\/uploads\/2023\/04\/Blooohggg.png\" alt=\"\" class=\"wp-image-836\" srcset=\"https:\/\/anchrom.in\/blogs\/wp-content\/uploads\/2023\/04\/Blooohggg.png 550w, https:\/\/anchrom.in\/blogs\/wp-content\/uploads\/2023\/04\/Blooohggg-480x215.png 480w\" sizes=\"(min-width: 0px) and (max-width: 480px) 480px, (min-width: 481px) 550px, 100vw\" \/><\/figure>\n\n\n\n<h1 class=\"has-medium-font-size wp-block-heading\"><a><\/a>HPTLC Method for <em>Cyperus rotundus<\/em> Rhizome<\/h1>\n\n\n\n<p class=\"wp-block-paragraph\"><a><\/a>HPTLC association has developed a method for <em>Cyperus rotundus<\/em> rhizome. This method also discriminates <em>C. rotundus<\/em> from other related species. This method is adopted in the International HPTLC Atlas for the identification of herbal drugs and in the British Pharmacopoeia, 2018. Standards isoeugenol and isoeugenyl acetate were used.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">USP method of <strong>ultrasonic extraction<\/strong> was used for standard and sample preparation with <strong>methanol <\/strong>as a <strong>diluent.<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">The plate layout was kept as per the parameters defined in USP chapter 1064. Mobile phase, toluene: ethyl acetate in the ratio <strong>9:1 v\/v<\/strong> was used for development.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">The development was carried out with <strong>controlled humidity<\/strong> of 33%. The plate can be automatically developed under controlled environmental conditions using <strong>CAMAG ADC2<\/strong> (Automatic development chamber).&nbsp;<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">After development, the plate was derivatized with anisaldehyde sulfuric acid reagent.
